General description
Sob-AM2 is a methyl amide derivative and a central nervous system (CNS)- selective prodrug of Sobetirome. Even in the absence of monocarboxylate transporter 8 (Mct8), Sob-AM2 can penetrate the brain and exhibit thyromimetic effects. It can promote extensive myelin repair in the brain and spinal cord of mouse models with demyelination. Sob-AM2 and other thyromimetics may also stimulate triggering receptor expressed on myeloid cell-2 (TREM2) expression in macrophages and microglia.

Thyroid : official journal of the American Thyroid Association, 28(9), 1211-1220 (2018-05-31)
Loss of function mutations in the thyroid hormone (TH)-specific cell membrane transporter, the monocarboxylate transporter 8 (MCT8), lead to profound psychomotor retardation and abnormal TH serum levels, with low thyroxine (T4) and high triiodothyronine (T3). Several studies point to impaired

Triggering receptor expressed on myeloid cells-2 (TREM2) is a cell surface receptor on macrophages and microglia that senses and responds to disease-associated signals to regulate the phenotype of these innate immune cells. The TREM2 signaling pathway has been implicated in

We have previously demonstrated that thyromimetics stimulate oligodendrocyte precursor cell differentiation and promote remyelination in murine demyelination models. We investigated whether a thyroid receptor-beta selective thyromimetic, sobetirome (Sob), and its CNS-targeted prodrug, Sob-AM2, could prevent myelin and axonal degeneration in
