General description

MTA1 gene is mapped to human chromosome 14q32.33.
Metastasis associated 1 (MTA1) is a 80kDa protein which possesses four domains and several interaction motifs. It is made up of 715 amino acid residues.

Specificity

Anti-MTA1 recognizes specifically MTA1.

Immunogen

synthetic peptide corresponding to amino acids 626-641 of human MTA1, conjugated to KLH via an N-terminal added cysteine residue. The immunizing sequence is not present in other members of the family, MTA2 and MTA3.

Application

Anti-MTA1 antibody produced in rabbit has been used in:
  • western blotting
  • immunohistochemistry
  • immunoprecipitation.

Biochem/physiol Actions

MTA1 interacts with cyclin-dependent kinase-activating kinase (CAK) is a component of the transcription factor II H (TFIIH) regulatory complex, suggesting its role as a signal transducer to mediate crosstalk between corepressor complexes and the general transcription machinery.
Metastasis associated 1 (MTA1) has been shown to be overexpressed in many types of cancers and has an important role in cancer malignancy. It takes part in the DNA repair process. As part of the nuclear remodeling and deacetylation complex (NuRD complex), it associates with deacetylated p53. MTA1 participates as a co-repressor and co-activator for many kinds of genes.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in frost-free freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilu-tions should be discarded if not used within 12 hours.
