General description

TRAF2 (TNF receptor associated factor 2) is one of the six members of the TRAF family. It is an adaptor protein, and is composed of a conserved TRAF domain in its C-terminal and a RING finger domain in its N-terminal. It also contains multiple zinc finger regions.

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. Biochem/physiol Actions

TRAF2 (TNF receptor associated factor 2) is recruited on activation of TNFR (tumor necrosis factor receptor) by TNF. It directly binds to TNFR2 and interacts with TNFR1 through TRADD (TNFR1-associated death domain protein). This protein activates JNK (c-jun N-terminal kinase) upon activation by ASK1 (apoptosis signal-regulating kinase 1). TRAF2 faciliates rapid vaccinia virus (VACV) entry and thus aids VACV replication. TRAF2 and RIP1 (receptor-interacting protein) up-regulation in leukocytes results in increased inflammatory responses in multiple sclerosis (MS) patients, and the elevation of this protein is seen in patients with relapsing-remitting multiple sclerosis (RRMS). The expression of this protein is linked with poor prognosis in activated B-cell (ABC)-like diffuse large B-cell lymphomas (DLBCL), where it activates NF-κB. It might also be the reason for chemoresistance in ABC-DLBCL.
