General description

Protein S100-A9 (UniProt P06702; also known as Calgranulin-B, Calprotectin L1H subunit, Leukocyte L1 complex heavy chain, Migration inhibitory factor-related protein 14, MRP-14, p14, S100 calcium-binding protein A9, S100A9) is encoded by the S100A9 (also known as 60B8AG, CAGB, CFAG, CGLB, L1AG, LIAG, MAC387, MRP14) gene (Gene ID 6280) in human. S100A9 is one of the 21 members of the S100 family of proteins. S100A8 (calgranulin-A), S100A9 (calgranulin-B) and the heterdimer S100A8/S100A9 (calprotectin) exert their immunological and inflammatory action via binding to TLR-4, RAGE and some other putative receptors. S100A8, calprotectin, and S100A9 are chemoattractants for neutrophils, and S100A9 is known to induce the expression and activation of beta2-integrins. S100A9 is also shown to stimulate neutrophil degranulation and phagocytosis, while both S100A8 and S100A9 can induce the secretion of inflammatory cytokines from human monocytes. In addition, S100A9 is shown to potentiate both fMLP- and GM-CSF-stimulated IL-8 production from neutrophils, although S100A9 on its own cannot induce neutrophil cytokine production in the absence of fMLP and GM-CSF.

Specificity

Clone 6B4 selectively detects human S100A9 with no cross-reactivity toward hS100A8, hS100A12, mS100A8, or mS100A9.

This mouse monoclonal Anti-S100A9 Antibody, clone 6B4, Cat. No. MABF854 is validated for use in Western Blotting and ELISA, for the detection of S100A9.
Western Blotting Analysis: A representative lot detected recombinant hS100A9 (rhS100A9), as well as endogenous hS100A9 in neutrophil extract and purified calprotectin (S100A8/S100A9 dimer), but not rhS100A8 & rhS100A12 or recombinant murine S100A8 & S100A9 (Dr Philippe Tessier, Laval University, Quebec, CA).
ELISA Analysis: A representative lot detected recombinant human S100A9 (rhS100A9) by direct/non-sandwich ELISA, but not rhS100A8 & rhS100A12 or recombinant murine S100A8 & S100A9 (Dr Philippe Tessier, Laval University, Quebec, CA).
ELISA Analysis: A representative lot detected recombinant human S100A9 (rhS100A9) as well as endogenous S100A9 in purified human calprotectin (S100A8/S100A9 dimer) by sandwich ELISA, but not rhS100A8 or rhS100A12 (Dr Philippe Tessier, Laval University, Quebec, CA).

Quality

Evaluated by Western Blotting in human spleen tissue lysate.

Western Blotting Analysis: 0.1 µg/mL of this antibody detected S100A9 in 10 µg of human spleen tissue lysate.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
