Target description

Nucleoprotein (UniProt: P07400; also known as Nucleocapsid protein, Protein N) is encoded by the N gene (Gene ID: 127897) in Lymphocytic choriomeningitis virus (LCMV). LCMV is a murine enveloped RNA virus of the arenavirus family. Its viral surface is densely covered with viral fusion glycoprotein spikes that form heterodimeric trimers, consisting of the peripheral receptor binding glycoprotein 1 (GP-1) and a transmembrane subunit (GP-2). The large segment of LCMV genome encodes for the RNA-dependent-RNA-polymerase (L) and the matrix protein (Z) that mediates viral assembly and budding. Its small segment (S) encodes the glycoprotein precursor (GPC) and the viral nucleoprotein (NP). GPC is then co-translationally cleaved by signal peptidase to produce a stable 58 amino acid stable signal peptide (SSP) and GPC that is post-translationally processed by the cellular site 1 protease to form GP-1 and GP-2. GP-1 is reported to mediate virion attachment to the cell surface followed by cell entry via receptor-mediated endocytosis. The GP-2 subunit is responsible for the pH-dependent fusion event in the acidic environment of the endosome to complete the virus cell entry process and release of virus ribonucleoprotein into the cell cytoplasm to initiate transcription and replication of the viral genome. The nucleoprotein of LCMV encapsidates the genome and protects it from nucleases. The encapsidated genomic RNA known as the nucleocapsid (NC) serves as template for viral transcription and replication. Clone VL-4 specifically reacts with LCMV nucleoprotein and does not exhibit reactivity with vaccinia or influenza virus infected cells. (Ref.: Urata, S., et al. (2018). 0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
