General description

14-3-3 protein beta/alpha (KCIP-1, Protein 1054, Protein kinase C inhibitor protein 1), epsilon (14-3-3E), eta (Protein AS1), gamma (KCIP-1, Protein kinase C inhibitor protein 1), theta (14-3-3 protein T-cell, 14-3-3 protein tau, Protein HS1), zeta/delta (KCIP-1, Protein kinase C inhibitor protein 1) are encoded by the YWHAB, YWHAE, YWHAH, YWHAG, YWHAQ, YWHAZ gene (Gene ID 7529, 7531, 7533, 7532, 10971, 7534, respectively) in human. Mammals express seven distinct 14-3-3 isoforms (gamma, epsilon, beta, zeta, sigma, theta, tau) that form multiple homo- and hetero- dimmers, thus rendering 14-3-3 as a key multifunctional regulatory molecule. 14-3-3 isomers have been implicated in several neurological disorders: for example isoforms gamma, epsilon and beta are expressed in the cerebrospinal fluid of Creutzfeldt-Jakob patients, but not patients suffering from other neurological conditions such as Alzheimer′s. The 14-3-3 pan antibody is one of a panel of antibodies that were generated against 14-3-3 isoforms and have demonstrated their potential for use as an ELISA-based diagnostic tool. These antibodies will allow the research to investigate the role of this protein in key cellular processes and neurological disorders such as Creutzfeldt-Jakob, Alzheimer′s and Parkinson′s disease.

Specificity

This antibody targets the N-terminal end sequences of 14-3-3 alpha/beta (YWHAB), gamma (YWHAG), zeta/delta (YWHAZ), epsilon (YWHAE), eta (YWHAH or YWHA1), and theta (YWHAQ).

Immunogen

A mixture of KLH-conjugated linear peptides corresponding the the N-terminal end sequences of human 14-3-3 alpha/beta (YWHAB), gamma (YWHAG), zeta/delta (YWHAZ), epsilon (YWHAE), eta (YWHAH or YWHA1), and theta (YWHAQ).

Application

Anti-14-3-3 pan, Cat. No. AB9748-I, is a highly specific rabbit polyclonal antibody, that targets 14-3-3 protein and has been tested in Immunohistochemistry (Paraffin), Immunoprecipitation, and Western Blotting and .
Immunohistochemistry Analysis: A 1:5,000 dilution from a representative lot detected 14-3-3 immunoreactivity in human prostate cancer, cerebellum, and tonsil tissue sections.

Immunoprecipitation Analysis: 5 µg from a representative lot immunoprecipitated 14-3-3 from 500 µg of A431 cell lysate.
