General description

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Histone H3 variants (H3.1, H3.2 and H3.3) have been implicated in the epigenetic memory of cellular state. Genome-wide patterns of H3 are dependent on amino acid sequence and change with cellular differentiation at developmentally regulated loci.

Specificity

Broad species cross-reactivity expected
Clone 3H10 recoginzes histone N-terminal peptide with Ser10 phosphorylation (pSer10) by bead-based peptide specificity assay. Clone 3H10 does not bind unphosphorylated peptides or peptides with only pThr11 or pSer28 modification.

Application

Anti-phospho-Histone H3 (Ser10) Antibody, clone 3H10, Alexa Fluor 555 is an antibody against phospho-Histone H3 for use in Immunocytochemistry.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected phospho-Histone H3 (Ser10) in HepG2 cells.
The unconjugated antibody (Cat. No. 05-806) is shown to be suitable also for peptide inhibition assay (PIA), flow cytometry, immunocytochemistry, immunofluorescence, multiplex assay, Western blotting applications.

Quality

Evaluated by Immunocytochemistry in HeLa cells.

Immunocytochemistry Analysis: A 1:100 dilution of this antibody detected phospho-Histone H3 (Ser10) in HeLa cells.

Target description

~17 kDa observed.

Purified mouse monoclonal IgG1κ antibody conjugate in PBS with 15mg/ml BSA and 0.1 % sodium azide.
