Gene Information

human ... PROK1(84432)

General description

EG-VEGF (endocrine gland-derived vascular endothelial growth factor) belongs to the prokineticin family. It is mainly expressed in the steroidogenic glands (ovary, testis, adrenal cortex and placenta). EG-VEGF is a small, secreted peptide and works as a tissue-specific pro-angiogenic mitogen and chemotactic agent. The gene is mapped to human chromosome 1p13.
Recombinant human EG-VEGF is a 9.6kDa protein consisting of 86 amino acid residues and ten cysteine residues that potentially form five pairs of intra-molecular disulfide bonds.

Biochem/physiol Actions

EG-VEGF (endocrine gland-derived vascular endothelial growth factor) is involved in angiogenesis, inflammatory responses and hematopoiesis. It also participates in female reproduction. EG-VEGF suppresses trophoblast invasion in pregnancy. Changes in the activity of this protein are associated with gestational complication, such as gestational trophoblastic diseases, repeated pregnancy losses, preeclampsia and intrauterine fetal growth restriction. EG-VEGF interacts with G protein-coupled receptors, prokineticin receptor 1 (PKR1) and prokineticin receptor 2 (PKR2). It is also involved in the pathology of Leydig-cell-tumors, papillary thyroid cancer, neuroblastoma, prostate cancer, gastrointestinal tumors, pancreatic ductal adenocarcinoma, Merkel cell carcinoma and multiple myeloma.

Physical form

Lyophilized with no additives.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.

Hematogenous metastasis, mainly hepatic metastasis, is a frequent metastatic mode in colorectal cancer involving angiogenic growth factors. Two angiogenic growth factors, in particular, Vascular endothelial growth factor (VEGF) and Prokineticin1(PROK1), are considered to have an important role in hematogenous metastasis
