General description

Strepavidin-collodial gold conjugate binds to molecules, such as primary antibodies, that have been previously conjugated with biotin.
Streptavidin is a protein produced by the bacterium Streptomyces avidinii. It has four binding sites for biotin as does avidin. Both have been used extensively as probes in immunochemical systems, conjugated to antibodies, enzymes, fluorochromes and gold. Avidin is highly basic (pI ~ 10.5), compared to streptavidin (pI ~ 5-6). Avidin has a slightly higher binding capacity for biotin than streptavidin does, but it has glycosylated side chains which give higher backgrounds in blotting techniques than does streptavidin, which is not a glycoprotein. Streptavidin is more resistant than avidin to dissociation into subunits by guanidinium chloride. Streptavidin may be conjugated to colloidal gold.

Application

For detection of biotinylated compounds.
Strepavidin-collodial gold conjugate is a component of streptavidin/colloidal gold-biotin detection systems use in immunoblotting, light microscopy, and immunoelectron microscopy applications. It is commonly used in postembedding immunocytochemistry.

Physical form

Suspension in 0.01 M phosphate buffered saline, pH 7.2, containing 1% BSA, 20% glycerol, and 15 mM sodium azide

  1. Can you provide details on how streptavidin is conjugated to gold nanoparticles? Specifically, I'd like to know if there is any covalent binding involved or if it's a passive binding process.

    1. Product S9059 was manufactured using passive conjugation of the STP with 10 nm gold colloid. It does not involve covalent or linker chemistry. The protein binds to the gold colloid through amino acid residues, including charge attraction (lysine), hydrophobic attraction (tryptophan), and sulfur bonding (cysteine).
