Human Xenograft Prostate

Cell Line Description

22Rv1 is a human prostate carcinoma epithelial cell line derived from a xenograft that was serially propagated in mice after castration-induced regression and relapse of the parental, androgen-dependent CWR22 xenograft. The cell line expresses prostate specific antigen (PSA). Growth is weakly stimulated by dihydroxytestosterone and lysates are immunoreactive with androgen receptor antibody by Western blot analysis. Growth is stimulated by epidermal growth factor (EGF) but is not inhibited by transforming growth factor β-1 (TGF β-1). This cell line is tumorigenic in nude mice.

Culture Medium

RPMI 1640 without phenol red (Sigma R7509) + 2mM Glutamine + 10% Foetal Bovine Serum (FBS)

Subculture Routine

Split sub-confluent cultures (70-80%) 1:5 to 1:10 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
