Application

Human AB serum is used in tissue engineering, transplantation and cell therapy applications for the expansion of mesenchymal stem cells (MSC) from adipose tissue or mesenchymal stromal cells from human bone marrow; for standardized limbal epithelial stem cell graft generation and transplantation; for ex vivo expansion of NK cells from peripheral blood in Hematopoeitic Stem Cell Expansion Medium; and for upgrading pretransplant human islet culture technology.
Human Serum has been used: as a component of phosphate-buffered saline with Tween® detergent (PBST) in bridging enzyme-linked immunosorbent assay (ELISA), as a mock biological sample for assessing the performance of paper-based ELISA assay with complex biological samples, as a diluent to create a dilution series of the baseline sample for the generation of standard curve for NISTmAb spiked into serum, as well as for multiple myeloma (MM) patient sample MM2232

Biochem/physiol Actions

Human AB serum is typically obtained by allowing whole blood to clot naturally, which is known as Off-The-Clot (OTC) serum. Human serum obtained from clotted whole blood is preferred as it usually contains higher levels and a more diverse range of growth factors for optimal cell growth. Human serum from blood-type AB donors lacks antibodies specific for both A and B blood-type antigens. This makes it a universal donor serum that is suitable for various cell culture types including human stem cell lines.

Packaging

Packaging may vary for different batches due to short supply of the bottles used to pack this product. The presence or lack of shrink wrap around the cap of the bottle doesn’t impact product integrity/sterility of the product.

It is not unusual for our human serum products to be cloudy. This product is from pooled serum and cloudiness indicates that lipids are present in the serum. No clear, lipid-free human serum products are available.

  1. Is this serum contain Uric acid? If yes how much, or how to check it?. If the serum is expired, is it okay to use?

    1. While uric acid is present in human serum, this product is not tested for uric acid levels. Should uric acid measurement is required, see product MAK483, Uric Acid Assay Kit, which is suitable for the determination of Uric acid in serum. It is not recommended to use products that are beyond the assigned expiration date.

  2. Once the product has been thawed, how should it be stored? How many times can be freezed and thawed?

    1. The magnesium levels are not determined for this serum product. The calcium levels are not tested on a lot-to-lot basis. However, historical information indicates calcium concentrations tend to range from 6.5 - 10 mg/dL.

    1. The immunoglobulin concentration of this product is not measured. This is a pooled product from adult AB negative males. Based on information published in the Geigy Scientific Tables, the average immunoglobulin levels for adult males, ages 20 – 70, are as follows:

      1.32 – 2.34 g/L
      IgM (not age dependent)
      0.77 g/L

  7. If this is "serum" derived from "plasma" the plasma would have contained anti-coagulant. To turn the plasma into serum it has to be clotted.  Has this product  been clotted to remove the fibrin? Or is it still in fact plasma?

    1. The product has been clotted. Blood is collected and treated with citrate (anticoagulant) which inhibits clotting initially. The blood is then centrifuged. When the spin is complete, layers will appear which allow a differentiation between the cells and the plasma. The plasma is removed and calcium is added to activate the clotting cascade. The preparation is then centrifuged again and the result is a large clot which is pelleted to the bottom of the tube with a serum layer on top.

    1. The use of human serum for a nucleic acid stability assay has not been performed as it is not part of our lab's product characterization focus. However, the citation titled "Improving gene silencing of siRNAs via tricyclo-DNA modification, PMID: 21687522" suggest that product H4522 may be suitable for this application.     1. Human Serum products H4522 and H6914 may not be suitable for complement assays when stored at -20°C due to potential degradation of complement activity.
