Specificity

Clone 2C20 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects MX1/MxA. It targets an epitope within 21 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal region of human MX1/MxA.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected MX1/MxA in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected MX1/MxA in lysates from A549 cells and 1:1,000 dilution in Human thymus tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MX1/MxA in Human thymus tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MX1/MxA in A549 cells.

Affinity Binding Assay: A representative lot of this antibody bound MX1/MxA peptide with a KD of 1.2 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Interferon-induced GTP-binding protein Mx1 (UniProt: P20591; also known as Interferon-induced protein p78, IFI-78K, Interferon-regulated resistance GTP-binding protein MxA, Myxoma resistance protein 1, Myxovirus resistance protein 1) is encoded by the MX1 gene (Gene ID: 4599) in human. MX1/MxA is a homotetrameric, interferon-induced dynamin-like GTPase that can oligomerizes into multimeric filamentous or ring-like structures by virtue of its stalk domain (aa 367-632). This oligomerization is critical for GTPase activity, protein stability, and recognition of viral target structures. Its C-terminal GTPase effector domain (GED; aa 574-662) is involved in oligomerization and viral target recognition. Its middle domain (aa 366-533) mediates self-assembly and oligomerization. It displays antiviral activity against a wide range of RNA viruses and some DNA viruses. Its target viruses include negative-stranded RNA viruses and HBV through binding and inactivation of their ribonucleocapsid. It is reported to antagonize reoviridae and asfarviridae replication and inhibit thogoto virus (THOV) replication by preventing the nuclear import of viral nucleocapsids. It differs from other small GTPases and heterotrimeric G proteins in features such as their large size (70-100 kDa), a relatively low affinity for GTP, and a high intrinsic rate of GTP hydrolysis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
