Specificity

Clone 1C3 is a ZooMAb^® rabbit recombinant monoclonal antibody that specifically detects KLF2. It targets an epitope within 20 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the N-terminal region of human KLF2.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell nuclear extract.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected KLF2 in HeLa cell nuclear extract.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution of this antibody detected KLF2 in Human stomach tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected KLF2 in MCF-7 cells.

Flow Cytometry Analysis: 0.1 g from a representative lot detected KLF2 in one million MCF-7 cells.

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected GST-tagged recombinant fragment corresponding to 56 amino acids from the N-terminal region of Human KLF2.

Affinity Binding Assay: A representative lot of this antibody bound KLF2 peptide with a KD of 9.2 x 10-8 in an affinity binding assay.

Target description

Krueppel-like factor 2 (UniProt: Q9Y5W3; also known as Lung krueppel-like factor, KLF2) is encoded by the KLF2 (also known as LKLF) gene (Gene ID: 10365) in human. KLF2 is a member of the krueppel C2H2-type zinc-finger protein family that serves as a transcription factor that binds to the CACCC box in the promoter of target genes and activates their transcription. It contains a nine amino acids transactivation domain (aa 43-51). Its expression is observed in mammalian development and is found in many different cell types. It plays a role in several biological processes including adipogenesis, embryonic erythropoiesis, epithelial integrity, inflammation, and T-cell viability. It is reported to inhibit proinflammatory activation of monocytes and KLF2 expression in monocytes is reduced by cytokine activation. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
