Specificity

Clone 2B4 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects ASH2. It targets an epitope within 19 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the C-terminal half of human Set1/Ash2 histone methyltransferase complex subunit ASH2.

Application

Quality Control Testing

Evaluated by Western Blotting in HL-60 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ASH2L in HL-60 cell lysate.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ASH2L in Jurkat cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected ASH2L in human colon tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound ASH2L peptide with a KD of 1.2 x 10-6 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Set1/Ash2 histone methyltransferase complex subunit ASH2 (UniProt: Q9UBL3; also known as ASH2-like protein) is encoded by the ASH2L (also known as ASH2L1) gene (Gene ID: 9070) in human. The Absent, Small, Homeotic discs 2-Like (ASH2L) is a transcriptional regulator that is a component of the Set1/Ash2 histone methyltransferase (HMT) complex composed of ASH2L, RBBP5, WDR5, HDPY-30, one of seven SET/MLL proteins (SETA, SETB, MLL1 to MLL5), and additional MLL complex-specific non-core proteins. This complex specifically methylates lysine 4 of histone H3. In association with RBBP5 and WDR5, it stimulates the histone methyltransferase activities of KMT2A, KMT2B, KMT2C, KMT2D, SETD1A and SETD1B. ASH2L is ubiquitously expressed in humans with the highest level of expression detected in the fetal liver and testis. Its expression is also observed in adult heart and testis. It plays a crucial role in mammalian development. Mice lacking ASH2L die prior to embryonic implantation. It can undergo monomethylation and dimethylation on arginine residues in the C-terminus with arginine 296 being the major site. However, methylation is not required for its nuclear localization and MLL complex integrity. Overexpression of ASH2L is reported to enhance p53-dependent apoptosis in response to chemotherapy. ASH2L contains a DNA-binding Forkhead-like DNA binding domain (aa 67-177) and an atypical PHD-type domain (aa 1-66) at its N-terminal region. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
