Application

La transferrina es la principal molécula sérica de unión al hierro y su liberación en la célula. Los sistemas de cultivo celular carentes de suero requieren un formato de liberación de hierro. La transferrina es la forma de administración de hierro preferente porque las células procesan el hierro ligado a la transferrina de una forma fisiológicamente adecuada a través de los receptores de transferrina de la superficie celular. La holo-transferrina humana es una transferrina de alta afinidad que puede utilizarse con muchos tipos de células. La holo-transferrina humana se carga con hierro antes de ser utilizada y puede añadirse directamente a medios de cultivo celular carentes de hierro o con poco contenido en hierro.
Se ha complementado la holo-transferrina humana en el Roswell Park Memorial Institute (RPMI) para análisis de proliferación de proeritroblastos[1] y también en medios de cultivo para espectroscopia de emisión con plasma intuitivamente acoplado.[2]

  2. What's the process of saturation? Do the cells express directly holo-Tf with iron? Or apo-Tf reacts with iron salts?

    1. The delivery of iron to cells by transferrins is a receptor-mediated and controlled process. Cells regulate the amount of iron they receive from the extracellular environment by varying transferrin receptor expression. Two iron-loaded transferrin molecules bind to one transferrin receptor. Once bound, they are transferred into acidic endosomes within the cell’s interior. At reduced pH, the iron is released and incorporated into intracellular proteins and the storage molecule ferritin. The apo-transferrin (iron-free) is recycled back to the exterior of the cell and released. Each transferrin receptor cycle can deliver up to four atoms of iron.

  3. Holo-Tf is described as "iron saturated", but there is a statement "Under normal circumstances, approximately one-third of transferrin iron-binding pockets are filled." I'm confused if the product I got (T0665) is 100% saturated with iron or not.

    1. This product is a Holo-Transferrin form which is generally considered to be iron-saturated. The specification for iron content of this product is 0.10 - 0.35%. Conversely, Apo-Transferrin is considered to be iron deficient. Apo-transferrin product T5391 has an iron content specification of <0.005%. Each Transferrin has the capacity to bind 2 iron (Fe3+) molecules. In a circulating system, the iron content is transitory and rarely (if ever) 100% saturated. This product is tested for total iron content. The number of binding sites that are filled is not measured.   4. What is the recommended holo-transferrin solvent and how should the dissolved protein be stored?

    1. This product is tested for solubility in water at 50 mg/mL. Rates are expected to be comparable when dissolving in an aqueous buffer or medium. Solutions are stable for 5-10 days at 2-8°C. This product has not been evaluated for long-term solution stability. However, medium supplements containing transferrin are stable when stored in aliquots at -20°C for up to one year.     1. We have found the information on transferrin in a reference book:The Plasma Proteins, F.W. Putnam, ed, volume 2, chapter 4, Table II.According to the table, Transferrin is 6 percent carbohydrate, citing G.A. Jamieson, J. Biol. Chem. 240(7), 2914-2920 (1965).The table also gives information on the percentage of each type of monosaccharide that is present.
