Inhibitor of 8-oxoguanine DNA glycosylase 1 (OGG1)
TH5487 is an inhibitor of 8-oxoguanine DNA glycosylase 1 (OGG1), which is part of the DNA base excision repair pathway. TH5487 as shown to inhibit OGG1-dependent proinflammatory gene expression by binding in the OGG1 active site, preventing OGG1 from engaging damaged DNA in vitro and in cells, resulting in decreased proinflammatory gene expression.

The onset of inflammation is associated with reactive oxygen species and oxidative damage to macromolecules like 7,8-dihydro-8-oxoguanine (8-oxoG) in DNA. Because 8-oxoguanine DNA glycosylase 1 (OGG1) binds 8-oxoG and because Ogg1-deficient mice are resistant to acute and systemic inflammation, we
