General description

Ankyrin repeat domain-containing protein 1 (ANKRD1) is a member of muscle ankyrin repeat protein. It comprises a nuclear localization signal, four repeats of an ankyrin motif, and a PEST-like sequence. The ANKRD1 gene is mapped to human chromosome 10q23.31. It is expressed in skeletal and cardiac muscles.
The protein encoded by this gene is localized to the nucleus of endothelial cells and is induced by IL-1 and TNF-alpha stimulation. Studies in rat cardiomyocytes suggest that this gene functions as a transcription factor. Interactions between this protein and the sarcomeric proteins myopalladin and titin suggest that it may also be involved in the myofibrillar stretch-sensor system. (provided by RefSeq)

Biochem/physiol Actions

Ankyrin repeat domain-containing protein 1 (ANKRD1) acts as a transcriptional regulator for cardiomyogenesis. It interacts with calsequestrin-2, intermediate filament protein desmin, and with Y-box transcription factor 1 (YB-1). Elevated expression of ANKRD1 is implicated in idiopathic dilated cardiomyopathy. The hepatitis C virus (HCV) coordinates with ANKRD1 for its entry and propagation. It serves as a co-activator of the p53 tumor suppressor protein and may regulate apoptosis in human ovarian cancer cells.
