General description

Pluronicpluronic® F-127, también conocido como poloxámero 407, es un copolímero no tóxico[1] y no iónico. Contiene óxido de polietileno-óxido de polipropileno dispuesto en una estructura tribloque para formar PEO-PPO-PEO simétrico.[2] Las propiedades tensioactivas de este polímero, derivadas de su naturaleza anfifílica, le permiten interactuar con superficies hidrófobas y membranas biológicas. [3]

Application

Pluronic® F-127 se ha utilizado:

  • como un sistema de administración de un péptido al sitio de la herida para estudiar el efecto del péptido en la cicatrización de las heridas en ratones.[4]
  • como revestimiento superficial para sellos de PDMS (polidimetilsiloxano) para volverlos repelentes a hidrogel.[5]
  • en la preparación de disolución indicadora de calcio de reserva para medir las actividades neuronales.[6]
  • como componente de la disolución salina tamponada con fosfato (PBS), para reducir la adhesión celular y proteica inespecífica a un dispositivo microfluídico basado en PDMS.[7]
  • Este copolímero tensioactivo no iónico es adecuado para utilizar como antiespumante en aplicaciones de cultivo de células de insectos.

Biochem/physiol Actions

Pluronic® F-127 es un gel termorreversible que encuentra su aplicación en diferentes industrias debido a su gran termosensibilidad, biodegradabilidad y biocompatibilidad.[1] Estas propiedades hacen que el polímero sea adecuado para utilizar como transportador versátil de medicamentos. Pluronic® F-127 mejora la eficiencia terapéutica de los medicamentos al aumentar su biodisponibilidad y su termoestabilidad.[8] Además, puede promover la unión celular y la formación de colágeno, lo que facilita la angiogénesis y la regeneración tisular. Debido a su eficacia, Pluronic F-127 se ha utilizado en la encapsulación de células madre mesenquimatosas (MSCs) para ayudar en la regeneración de tejidos con bajos niveles de vascularidad, como los tejidos epiteliales, los tejidos adiposos, los cartílagos, los tendones y los tejidos óseos.[1]

    1. The polydispersity is not a test specification for this product. The product is an ABA-type copolymer of poly (ethylene oxide) (PEO=A) and poly (propylene oxide) (PPO=B), where the A and B blocks have the following values: A = 101, B = 56. The approximate relative amount of PEO in the poloxamer is ca. 70% and the average molecular weight of the PPO is 4000. The molecular structure is shown below:

    1. Pluronic F-127 will form gels above 10 °C when used at concentrations of 18-50%. It will re-liquefy when cooled to below 10 °C. The gel should remain stable during autoclaving.
