Descripción general

Inorganic sulfate is one of the most abundant anions in mammalian plasma. Sulfate plays important physiological roles in activating and detoxifying xenobiotics, steroids, neurotransmitters, and bile acids.[1] Sulfate is needed for the biosynthesis of cerebroside sulfate, glycosaminoglycans, and heparin sulfate. Undersulfation of cartilage proteoglycans has been associated with human inherited osteochondro-dysplasia disorders.[2] In mammals, sulfate homeostasis is regulated by the kidney. The majority of filtered sulfate is absorbed in the proximal tubules and only 5-20% of the filtered load is excreted into the urine.[3][4]

Principio

The kit employs an improved method utilizes the quantitative formation of insoluble barium sulfate in polyethylene glycol. The turbidity measured at 600 nm is proportional to sulfate level in the sample.

  1. Can the kit be applied to automated biochemical analysers?

    1. This kit has not been specifically tested with automated biochemical analyzers. The current protocol is designed for manual use; however, there is no indication that the kit cannot be used with automated biochemical analyzers.

  2. Can the MAK132 Sulfate assay kit be used to test wine instead of plasma? If so, are there any recommended protocol adjustments for this?

    1. The MAK132 assay kit is designed for testing sulfates, not sulfites, which are more commonly associated with wine. There is no available data on this kit being previously tested with wines. If the wine sample is diluted and/or the color is reduced through treatment with carbon or polyvinylpyrrolidone (PVPP) and the sample sulfate levels are within the detection range of 0.02 mM (0.19 mg/dL) to 2 mM (19.2 mg/dL) in a 96-well plate assay, then it might work for testing wine instead of plasma.
