General description

Interleukin-2 (IL-2) is a cytokine produced by activated T cells in response to antigen stimulation. IL-2 is important for proliferation of lymphocytes, natural killer cells and macrophages and in the differentiation of CD4+ cells into Th1 and Th2 effector cells. The effects of IL-2 are mediated by the signalling activated by its binding to IL-2 receptor (IL-2 R). IL-2 R which contains three membrane subunits: α, β and γ. The β and the γ subunits complex results in the binding of IL-2 with higher affinity. IL-2 R β is expressed on resting monocytes and B cells, CD8+ and CD4+T cells, NK cells and neutrophils and mature thymocytes. IL-2 R activates key signalling pathways such as MAPK, JAK/STAT as well as PI3K/Akt. Activation of these pathways modulates production of inflammatory cytokines, survival and progression through cell cycle
Anti-Human IL-2 soluble receptor β (IL-2 sRβ) specifically reacts with human IL-2 Rβ. The antibody shows less than 15% cross reactivity with recombinant human IL-3 sRα, less than 5% cross reactivity with recombinant human IL-1 RII, IL-2 sRγ, IL-5 sRα, and less than 2% cross reactivity with recombinant human IL-2 sRα, IL-4 sR, and IL-5sRβ. In addition, the antibody shows no cross reactivity with other cytokines tested.

Specificity

The antibody will neutralize human cell surface IL-2 receptor β mediated-bioactivity. For cells expressing high affinity IL-2 receptors, use of both anti-IL-2 Rα and anti-IL-2 Rβ is recommended. The antibody shows ≤15% cross-reactivity with recombinant human IL-3 sRα, ≤5% cross-reactivity with recombinant human IL-1 RII, IL-2 sRγ, IL-5 sRα, and ≤2% cross-reactivity with recombinant human IL-2 sRα, IL-4 sR, and IL-5 sRβ.

Immunogen

recombinant human IL-2 soluble receptor β (IL-2 sRβ) expressed in Sf 21 insect cells.

Application

Anti-Human IL-2 soluble receptor β (IL-2 sRβ) antibody may be used for immunoblotting at a working concentration of 1-2 μg/ml. For indirect ELISA, the recommended concentration is 0.5-1.0 μg/ml. The antibody is suitable for IL-2 sRβ neutralization reactions at a working antibody concentration of 10-30 μg/ml.
