Gene Information

General description

FRK (fyn-related kinase) is a tyrosine kinase, with a molecular weight of 54kDa. It contains SH2 and SH3 (src homology) domains. It is predominantly expressed in tissues of epithelial origin such as, liver, kidney, breast and colon. Its SH2 domain contains a putative bipartite nuclear localization signal.

Biochem/physiol Actions

FRK (fyn-related kinase) controls growth by phosphorylating perinuclear proteins. It inhibits cell growth both in a pRb (retinoblastoma)-dependent and independent manner. It cause cell cycle arrest at G1 phase in breast cancer cells. Certain mutations in this gene lead to its constitutive activation, as seen in hepatocellular adenomas. Its up-regulation in gliomas prevents both invasiveness and migration of glioma cells. This is attributable to its promotion of N-cadherin/β-catenin complex formation.

Physical form

Supplied in 50 mM Tris-HCl, pH 7.5, with 150 mM NaCl, 0.2 5mM DTT, 0.1 mM EGTA, 0.1 mM EDTA, 0.1 mM PMSF, and 25% glycerol.

Hepatocellular adenomas (HCA) are benign liver tumors predominantly developed in women using oral contraceptives. Here, exome sequencing identified recurrent somatic FRK mutations that induce constitutive kinase activity, STAT3 activation, and cell proliferation sensitive to Src inhibitors. Fyn-related kinase (FRK), a member of Src-related tyrosine kinases, is recently reported to function as a potent tumor suppressor in several cancer types. Our previous study has also shown that FRK over-expression inhibited the migration and invasion of glioma cells.
