Application

Amphotericin B is a polyene antifungal antibiotic from Streptomyce sp. It has a high affinity for sterols, especially ergosterols, of both fungal and bacterial cell membranes. Normal usage in maintenance of antifungal cell cultures is 2.5 mg/L with penicillin and streptomycin used in the medium.

La anfotericina B se une a los esteroles formando poros en la membrana y provocando la salida de moléculas pequeñas. La máxima eficacia del producto es contra hongos y levaduras.

Packaging

Solubilized Amphotericin B is packaged in amber glass bottle.

Preparation Note

Amphotericin B is soluble at 19.6-20.4 mg/ml in water, at natural pH without heating, giving a yellowish clear to slightly hazy solution.
Aqueous solutions cannot be sterile filtered due to poor solubility. If reconstituted in sterile water, there is no need to filter sterilize.

It is also soluble in DMSO (30-40 mg/mL) and in dimethylformamide (2-4 mg/mL).

Other Notes

After reconstitution with water, the product will be a suspension of Amphotericin B, containing deoxycholate as the solubilizing agent. The lyophilized formulation is ~45% Amphotericin B and 35% sodium deoxycholate in sodium phosphate and sodium chloride.
Keep container tightly closed in a dry and well-ventilated place. Product is sensitive to light and moisture. Keep in a dry place.
The actual solid weight of the packaged product is calculated according to the UV purity.

Disclaimer

Amphotericin B remains active for 3 days in culture at 37°C. At -20°C, and with protection from air and light, the product remains active for 5 years.

    1. This is a powdered product. The potency in solution is dependent upon the concentration at which it is prepared. The minimum potency specification for this product is 240 µg/mg.     1. Amphotericin B is soluble in DMSO at 30-40 mg/mL. Add 100 mg powder to 40 mL DMSO in order to achieve a 2.5 mg/mL concentration.

We recommend to use Product No. A9528 at 5.6 mg solid per Liter.  Cell culture tested amphotericin B solubilized preparation is approximately 45% amphotericin B, 35% sodium deoxycholate, with the balance being sodium phosphate. The amphotericin B is at least 80% amphotericin B and 3-5% amphotericin A by HPLC.  The exact amount of amphotericin B in the product can be found on the lot specific Certificate of Analysis under Potency.
