General description

Recombinant, mouse sclerostin (SOST) consisting of amino acids 24-211, expressed in NS0-derived murine myeloma cells with an N-terminal His•Tag. SOST is expressed by osteoclasts in developing bones of mouse embryos and acts as a negative regulator of bone formation through repression of BMP-induced osteogenesis.

Biochem/physiol Actions

Inhibition Assay: Inhibits hBMP-6-induced alkaline phosphatase by 50-80% in MC3T3-E1 mouse osteoblastic cells when used at a concentration of 30 µg/ml.

Toxicity: Standard Handling (A)

Physical form

Lyophilized from 0.2 µm filter-sterilized PBS, BSA.

Reconstitution

Following reconstitution refrigerate (4°C) for short-term storage or aliquot and freeze (-20°C) for long-term storage. Stock solutions are stable for up to 1 month at 4°C or for up to 3 months at -20°C.
Reconstitute to 200 µg/ml in sterile PBS containing at least 0.1% human or bovine serum albumin.

Mechanical loading is essential for the maintenance of musculoskeletal homeostasis. Cartilage has been demonstrated to be highly mechanoresponsive, but the mechanisms by which chondrocytes respond to mechanical stimuli are not clearly understood. The goal of the study was to determine
