General description
GSTP1 (glutathione S-transferase P) belongs to the superfamily of glutathione-S-transferases. It is widely expressed and is mainly present in the lung, esophagus and placenta. This gene is mapped to human chromosome 11q13.
Application
GSTP1 human has been used as a standard to measure GST (glutathione S-transferase) activity.
Biochem/physiol Actions
Members of GST (glutathione S-transferase) superfamily are responsible for the metabolism and detoxification various carcinogens, toxins and drugs. GSTP1 can associate with many kinases. Under normal conditions, it associates with c-Jun-N-terminal kinase (JNK) and c-jun. In presence of oxidative or chemical stress, it dissociates from JNK, resulting in its activation. It also associates with peroxiredoxin 1, thereby reactivating it. It interacts with tumor necrosis factor receptor-associated factor 2 (TRAF2). This interaction suppresses TRAF2-associated activation of apoptosis signal-regulating kinase 1, JNK and p38. GSTP1 is also a Cdk5 (cyclin-dependent kinase 5) regulatory protein and determines Cdk5 activity in Alzheimer′s disease brains, as well as in various cancer and neuronal cells. Mutations in the GSTP1 gene might be associated with the severeness of airway dysfunction, treatment outcome of advanced NSCLC (non-small cell lung cancer) in patients and increased risk of OLK (oral leukoplakia).
Physical form
0.5 mg/mL solution in 20 mM Tris-HCl buffer (pH 7.0) containing 1 mM EDTA, 0.1 mM PMSF and 30% glycerol.
