General description

Lipid peroxidation is the degradation of lipids that occurs as a result of oxidative damage and is a useful marker for oxidative stress. Polyunsaturated lipids are susceptible to an oxidative attack, typically by reactive oxygen species, resulting in a well-defined chain reaction with the production of end products such as malondialdehyde (MDA). Lipid peroxidation may contribute to the pathology of many diseases including atherosclerosis, diabetes, and Alzheimer′s.

Principle

In this kit, lipid peroxidation is determined by the reaction of MDA with thiobarbituric acid (TBA) to form a colorimetric (532 nm)/fluorometric (λex = 532/λem = 553 nm) product, proportional to the MDA present.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   3. Is the MAK085-1KT Lipid Peroxidation (MDA) Assay Kit compatible with tissue samples stored in 4% paraformaldehyde PBS?

    1. The assay has not been tested in PFA-fixed or methanol-fixed samples, and it is not predicted that the kit would work robustly or consistently in these samples.

  4. Is MAK085 suitable for use on plant samples?

    1 answer
    1. The MAK085 kit has never been tested on plant samples, and thus there is no optimized protocol for plant samples. This kit has only been validated for tissue, cells, serum, or plasma samples. It is possible to adapt the protocol for plant samples and test it, but there is no guarantee that it will work.

  5. We are planning an MDA assay using mouse tissue. Should I avoid cryopreservation of the samples? How should we store the samples if they cannot be assayed immediately?

    1. Whenever possible, It is recommended to use fresh samples. If sufficient fresh samples are not available, samples may be flash frozen in liquid N2 and stored at -80C. A storage time frame has not been determined. Cryopreservation of samples should be avoided as this may affect the integrity biomolecules and contribute to oxidative stress.
