General description

The kit contains all reagents and solutions required for determining superoxide dismutase activity in an indirect assay method based on xanthine oxidase and a novel color reagent. The chemical and biochemical properties of the color reagent used in this kit guarantee a very convenient application and linearity of test results over a broad range.

Application

SOD Assay Kit has been used for the determination of superoxide dismutase activity in mammalian cells.[1][2][3]

Biochem/physiol Actions

Superoxide dismutases are enzymes responsible for the elimination of superoxide radical (O2-), thereby protecting cells from free radicals-associated damages.[4] They catalyze the dismutation of two superoxide radicals to hydrogen peroxide and oxygen.[5]

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   3. Hello, Please what is the quantity of cell sample is required for use in this kit ?

    1. The recommended number of cells is 9x10*6 for adherent lines. Leukocytes have been tested successfully at 1.2x10*7 cells. Please see the link below to review the product datasheet:

  4. Can we use that kit without having SOD? And the shiping can be done at room temperature and put in fridge after reception?

    1. The superoxide dismutase (SOD) is required for preparation of an inhibition curve. This is a SOD inhibitory assay kit. See SOD product S7571, which is suggested for use with this kit. This kit is shipped on wet ice to preserve the quality of the transit. Upon receipt, the item should be stored at 2-8°C. If the interest is in calculating SOD activity in a sample, product CS0009 may be a kit of interest. See the links below to review additional information.

  5. Hello, Please what is the quantity of tissue sample is required for use in this kit

    1. This product is not routinely tested for use with solid tissue samples. However, the following general preparation method for 100 mg of tissue is recommended:
      1. Rinse the tissue with saline solution to remove residual blood, then gently dab the tissue with paper towels before proceeding to weigh it.
      2. Add 400-900 ml of sucrose buffer (0.25 M sucrose, 10 mM Tris, 1 mM EDTA, pH 7.4) and use a Teflon homogenizer to process the sample. If needed, subject the homogenized sample to sonication on an ice bath (60W with 0.5 second intervals for 15 min).
      3. Centrifuge the homogenized sample at 10,000 g for 60 min at 4ºC then carefully transfer the resulting supernatant to a new tube for measurement.

  6. What is the SOD concentration?

    1. As per the protocol of 19160: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/product/documents/254/301/19160dat.pdf, the kit doesn't include SOD standard, that is to say, the SOD standard needs to be purchased separately. As shown on page 2 of the protocol, if assay monitoring is required, the SOD standard should be diluted in the range of 0.001-200 U/mL, indicating the detection range of the kit is 0.001-200 U/mL.

    1. Dilute SOD with Dilution Buffer to prepare SOD standard Solutions if necessary for assay monitoring as follows:  200 U/mL, 100 U/mL, 50 U/mL, 20 U/mL, 10 U/mL, 5 U/mL, 1 U/mL, 0.1 U/mL, 0.05 U/mL, 0.01 U/mL, and 0.001 U/mL.
