Application

Phthaldialdehyde Reagent has been used for the derivatization plasma samples prior to high performance liquid chromatography.[1]
Precolumn derivatization reagent for primary amines and amino acids. The fluorescent derivative can be detected by reverse-phase HPLC. The reaction requires OPA, primary amine and a sulfhydryl. In the presence of excess sulfhydryl, amines can be quantitated. In the presence of excess amine, sulfhydryls can be quantitated.
Useful for investigators requiring an alternate sulfhydryl moiety. Requires addition of desired sulfhydryl compound for measuring amines. Requires addition of amine to measure sulfhydryls.

Linkage

Same as P 0532, but contains no 2-mercaptoethanol.

Questions

1–2 of 2 Questions
  1. Can you provide a more concrete concentration than what is listed on the SDS?

    1 answer
    1. The concentration is 1 mg o-phthaldialdehyde per mL solution.

      Helpful?

  2. What is the difference between Solution incomplete & Solution complete? Is there any effect on protein derivatization if 2-mercaptoethanol is missing? What is the main effect of 2-mercaptoethanol on the derivatization of amino acids?

    1 answer
    1. The formulation for P0532 is Brij 35, o-phthaldialdehyde, methanol, 2-mercaptoethanol, potassium hydroxide, and boric acid.
The formulation for P7914 is Brij 35, potassium hydroxide, and boric acid. It does not contain the 2-mercaptoethanol found in the complete reagent solution, P0532.
While it is possible to use OPA without 2-mercaptoethanol, it is often needed to ensure that disulfide bonds are cleaved. Without this cleavage, a complete reduction of primary amines may not occur. The OPA incomplete, P7914, is offered to allow for the addition of fresh 2-mercaptoethanol for each derivatization. This component can degrade over time, particularly if stored improperly.
See the link below for the product information sheet of the complete version, P0532. Page 2, Figure 1B, demonstrates the effects of degraded 2-mercaptoethanol.
