Application

Murashige and Skoog Basal Medium has been used:
  • to germinate the seeds for seedling growth and germination studies
  • in the suspension culture of Nicotiana plumbaginifolia for transformation
  • in the root culture of hairy clone A. artemisiifolia T4

Preparation Note

Formulated to contain 4.4 grams of powder per liter of medium.
Murashige and Skoog medium can be reconstituted from powder or by combining products that are major components of complete M&S medium, such as macronutrient, micronutrient and vitamin mixtures.
Murashige and Skoog medium (M0404) contains the macro- and micronutrients of M&S basal medium as described by Gamborg, et al (1968). It can be supplemented with organics and with auxins (IAA) and cytokinins (Kinetin) to generate a complete medium for growth plant tissue culture.
With the macro- and micronutrients as described by Murashige and Skoog (1962) and the vitamins as described by Gamborg, et al. (1968).

  1. The purchase options are "1 liter" and "10 liter" pack sizes. Does that mean enough powder with which to make 1 liter / 10 liters of finished media (4.4 g / 44 g of powder), or does it mean 1 l / 10 l of powder.

    1. The -1L pack size will contain enough powder (4.4g) to make 1L of finished media. The -10L pack size will provide enough powder for 10L of finished media.

  2. What is electric conductivity of Murashige-Skoog culture solution?

    1. The Murashige and Skoog media products have not been tested for conductivity.

  3. I would like to know if there is any problem in irradiating with UV light, media culture prepared with the medium.

    1. Testing protocols for plant culture media include sterilization by autoclaving. Heat-labile components are filter-sterilized and added after autoclaving, prior to asceptic dispensing to sterile vessels. UV sterilization has not been investigated as a sterilizing technique for culture media products, although this does appear to be growing in popularity. Keep in mind that UV would not melt agar used in solid media formats. Autoclaving or boiling would still be required.
