General description

The gene HSD17B4 (17-β-hydroxysteroid dehydrogenase 4) is mapped to human chromosome 5q23.1. It is also referred to as D-bifunctional protein (DBP). The protein contains an amino-terminal 3-hydroxyacyl-CoA dehydrogenase domain, a central 2-enoyl-CoA hydratase domain and a carboxyl-terminal sterol carrier protein 2-like domain. After transfer to peroxisomes, the protein is proteolytically cleaved to a dehydrogenase unit and a hydratase unit.

Biochem/physiol Actions

HSD17B4 (17-β-hydroxysteroid dehydrogenase 4) is a multifunctional peroxisomal enzyme. It is mainly involved in fatty acid β-oxidation and steroid metabolism. Mutations in HSD17B4 result in ovarian dysgenesis, hearing loss and ataxia of Perrault syndrome. In infants, mutations in HSD17B4 cause chylous ascites, polyhydramnios, claw hands and hammer toes.
