General description

Dodecylphosphocholine is a structural analog of lauroyl lysophosphatidylcholine that is more stable to hydrolytic degradation. The lipid is often used as a detergent for membrane protein solubilization and purification.Dodecylphosphocholine has been shown to improve paracellular permeability of hydrophilic compounds by modulating the tight junctions. This may have implications on its use for oral or nasal drug delivery.
MAPCHO-12 or n-dodecylphosphocholine is a detergent with short chain.

Application

MAPCHO-12 is suitable for use as detergent for B. subtilis cytochrome b558 protein for nuclear magnetic resonance experiments. It is also suitable for use in the purification of cytoplasmic tail of gp41 (gp41CT) protein and micelle preparation.

Biochem/physiol Actions

MAPCHO-12 or n-dodecylphosphocholine in combination with 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) is used for membrane-mimetic bicelle system for orientation based studies in membrane proteins. It provides a zwitterionic surface in the membrane micelle structure.

The intestinal epithelium is a significant barrier for oral absorption of hydrophilic drugs because they cannot easily traverse the lipid bilayer of the cell membrane and their passage through the intercellular space (paracellular transport) is restricted by the tight junctions.
