Specificity

Clone 1H2 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects BRCA2. It targets an epitope within 22 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal region of human BRCA2.

Application

Quality Control Testing

Evaluated by Western Blotting in Human brain tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected BRCA2 in Human brain tissue lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected BRCA2 in Human cerebral cortex, Human breast cancer tissue, and normal Human breast tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound BRCA2 peptide with a KD of 2.7 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected BRCA2 in A431 and MCF-7 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Breast cancer type 2 susceptibility protein (UniProt: P51587; also known as Fanconi anemia group D1 protein, BRCA2) is encoded by the BRCA2 (also known as FACD, FANCD1) gene (Gene ID: 675) in human. BRCA genes (1 and2) produce proteins that help repair damaged DNA. BRCA2 is involved in double-strand break repair and/or homologous recombination. It binds RAD51 and potentiates recombinational DNA repair by promoting assembly of RAD51 onto single-stranded DNA (ssDNA). It acts by targeting RAD51 to ssDNA over double-stranded DNA, enabling RAD51 to displace replication protein-A (RPA) from ssDNA and stabilizing RAD51-ssDNA filaments by blocking ATP hydrolysis. BRCA2 is phosphorylated by ATM upon irradiation-induced DNA damage. Phosphorylation by check point kinases 1 and 2 regulates its interaction with RAD51. Phosphorylation at serine 3291 by CDK1 and CDK2 is reported to be low in the S phase when recombination is active but increases as cells progress towards mitosis. This phosphorylation prevents homologous recombination-dependent repair during S phase and G2 by inhibiting RAD51 binding. Mutations in BRCA2 gene are known to increase susceptibility to breast and cervical cancers. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
