Application

SPARC has many useful properties including inhibiting cell spreading and diminishing focal contacts in vitro, abrogating the effects of PDGF and beta-FGF, binding directly to several components of the ECM, and undergoes proteolysis that exposes and releases regions in the protein that correlates with biological activities.

Biochem/physiol Actions

SPARC is expressed in many tissues including bone, skin, skeletal muscle, reproductive tissues, and embryonic connective tissue. It is expressed in high levels in tissues that are undergoing morphogenesis, remodeling and repair.

Components

SPARC, or Secreted Protein Acidic and Rich in Cysteine, is a 283-287 AA Ca2+ binding to glycoprotein with an apparent molecular weight of 40-43 kDa.

Caution

The lyophilized powder should be stored frozen at -20°C. The reconstituted product should be stored frozen at -70°C.

Preparation Note

This product is supplied as a lyophilized powder from phosphate buffered saline at 25 μg protein/vial. It should be reconstituted aseptically with sterile water, producing a solution of 0.1 to 0.3 mg protein per mL. SPARC is initially purified from mouse parietal yolk sac cells under conditions that preserve biological activity.
