General description

Monoclonal Anti-Nup98 (rat IgG2c- isotype) is derived from the hybridoma 2H10 produced by the fusion of mouse myeloma cells (SP2 cells) and splenocytes from WKY/NCrj rat immunized with a recombinant fragment of human Nup98. Nup96 and Nup98 are the nucleoporins that are formed by the cleavage of 186 kDa precursor protein.
Nucleoporin-98kDa (Nup98) is part of the nuclear pore complex. It possesses two domains and the gene encoding it is localized on human chromosome 11.

Application

Anti-Nup98 antibody, Rat monoclonal may be used in:
  • enzyme linked immunosorbent assay (ELISA)
  • immunoblotting
  • immunocytochemistry

Biochem/physiol Actions

Nucleoporin-98kDa (Nup98) is involved in the translocation of RNA and proteins across the nucleus. It is also a regulator of chromosomal segregation and gene expression. The protein also binds to other nucleoporins and is associated with acute myelogenous leukemia.
Nup98 functions as a docking protein for cytosol mediated docking of import substrates. Securin protein damage caused by the anaphase-promoting complex (APC) during mitosis, is regulated by the nucleocytoplasmic transport factors Rae1 and Nup98. In mice, collective haploinsufficiency of Rae1 and Nup98 gene causes premature separation of sister chromatids and severe aneuploidy. In addition, it also leads to untimely degradation of securin. Chromosome rearrangement between the HOXA9 (homeobox A9) gene and the nucleoporin gene Nup98 has been observed in some human myeloid leukemia patients.

The nucleoporin Nup98 is an essential component of the nuclear pore complex. This peripheral nucleoporin with its Gly-Leu-Phe-Gly (GLFG) repeat domain contributes to nuclear-cytoplasmic trafficking, including mRNA export. In addition, accumulating studies indicate that Nup98 plays roles in several important

NUP98 is a recurrent fusion partner in chromosome translocations that cause acute myelogenous leukemia. NUP98, a nucleoporin, and its interaction partner Rae1, have been implicated in the control of chromosome segregation, but their mechanistic contributions to tumorigenesis have been unclear.
