General description

Epidermal growth factor receptor substrate 15 (EPS15) localizes to the clathrin-coated pits at the plasma membrane. It contains three N-terminal EPS15 homology (EH) domains, an intermediate coiled-coil domain and a binding domain at the C-terminal end.

Application

Biochem/physiol Actions

Epidermal growth factor receptor substrate 15 (EPS15) functions in the endocytosis and down regulation of growth factor (GF) receptors, which is very important in GF signaling. It interacts with the endosomal sorting protein: hepatocyte growth factor–regulated tyrosine kinase substrate (Hrs) and is associated with the clathrin-coated pit adaptor protein 2 (AP2) complex. It functions in the upstream process of endocytosis. The domains of EPS15 are involved in the stimulation of clathrin assembly at newly forming clathrin coated pits through their interaction with the adaptor protein AP180. They are also needed for the recruitment of Eps15 to clathrin-coated pits.
