General description

Heme is a small molecule present either free or bound to hemoglobin in the bloodstream of mammals. This is an alternative source of iron within the host that contains a porphyrin ring containing a Fe2+ ion called hemin. [1]

Application

Hemin was used to study conjugation, characterization and toxicity of lipophosphoglycan-polyacrylic acid conjugate for vaccination against leishmaniasis.[2]

    1. This product is tested for solubility at 25 mg/mL in 1.4 M NH4OH. According to a published article, this material is also soluble at 50 mg/mL in 1.0 M NaOH. Please see the link below to review this publication:

  2. What is the recommended method of sterilization in a culture medium with Hemin addition? Autoclaving at 112°C or filter sterilization?

    1. Stock solutions of Hemin are typically added to medium and autoclaved at 121 °C for 15-20 minutes. Stock solutions may also be filter sterilized and added aseptically to sterile medium.

  3. I couln´t understand when you say: "There is no shelf life date assigned to this product. The guarantee of quality is for one year from the time of shipment for bottles stored as recommended." Then for Hemin - from bovine we will not have a Exp.date?

    1. A number of products are not included in expiration testing programs, as there is no indication to conclude they are unstable. The only date shown on the certificate of analysis will be the Release Date. For these products there is no stated Retest, Expiration or Use-by-Date Date. We recommend handling per defined conditions as printed in our product literature and website product descriptions. Products should be routinely inspected by the customer to ensure they perform as expected. For those products, only our standard warranty of 1 year from the date of shipment of products is applicable. For further information on our warranty please refer to terms and conditions of sale.

    1. Sigma-Aldrich has not evaluated the solution stability of hemin solutions.  Also, we have not found any information on solution storage in the literature. We recommend that solutions be prepared fresh.

  5. What are some applications for Hemin, Product H9039?

    1. Hemin-agarose has reportedly been prepared for use in affinity chromatography of heme-binding proteins.  See Tsutsui, K., Affinity chromatography of heme-binding proteins: Synthesis of hemin-agarose. Meth. Enzymol. 123, 331, (1986). Sigma supplies a hemin agarose, product H6390. Hemin can also be used as a component in culture media for the cultivation of bacteria, such as Leishmania tarentolae.
