General description

TRIM33, also known as E3 ubiquitin-protein ligase TRIM33, Ectodermin homolog, RET-fused gene 7 protein, Rfg7, Transcription intermediary factor 1-gamma, TIF1-gamma, or Tripartite motif-containing protein 33, and encoded by the gene TRIM33/KIAA1113/RFG7/TIF1G, is an E3 ubiquitin protein ligase. Among its targets is SMAD4 for which ubiquitination facilitates its nuclear exclusion and degradation. SMAD3 is also a target for ubiquitination, and together, TRIM33′s association promotes erythroid differentiation of hematopoietic stem cells. Finally mono-ubiquitination of various targets by TRIM33 affects their ability to form complexes which often hampers their activity and thus regulates their subsequent actions. Such is the case of SMAD4 and its interaction with TGF-beta/BMP signaling. Monoubiquitination of SMAD4 by TRIM33 hampers its ability to form a stable complex with activated SMAD2/3 resulting in inhibition of TGF-beta/BMP signaling cascade. TRIM33 is localized in nucleus in nuclear dots or bodies and is expressed in most tissues and stem cells during development as well as in the adult in multiple tissues.
