General description

The COL3A1 gene mapped to human chromosome 2q32.2, encodes the alpha 1 chain of type III collagen. Type III collagen is an extracellular matrix protein containing a long triple-helical domain made of three alpha 1 chains. Collagen III is a major structural component in hollow organs such as large blood vessels, the uterus and the bowel. Human type III collagen is purified by serial salt precipitations, alcohol precipitation, and DEAE chromatography of an acetic acid extraction of human placenta, following mild pepsin digestion. Product Source: Human placenta, negative for HbsAG, HCV and HIV 1 & 2 antibodies.

Application

Human Collagen Type III has been used:
  • as an extracellular matrix (ECM) protein in microarray analysis, to coat nitrocellulose film slides for probing and detection experiments
  • to coat microfluidic migration devices and to study the impact of extracellular matrix (ECM) on cell migration
  • as a positive control in Gpr56 PRESTO-Tango assay and GTP-Rho pull-down assay, to evaluate its effects on cellular signaling and interactions

Biochem/physiol Actions

Collagen III is initially produced as procollagen, a protein consisting of three pro-alpha1(III) chains that form the triple-stranded, rope-like molecule. After being synthesized, the procollagen molecule is modified by the cell. The type III procollagen molecules are released from the cell and are processed by enzymes that clip small segments off either end of the molecules to form mature collagen. The mature collagen molecules assemble into fibrils. Cross-linking between molecules produces a very stable fibril, contributing to collagen tissue strengthening function

Liquid in 0.5 M acetic acid, pH 2.5.

Storage and Stability

Maintain at -20ºC in undiluted aliquots for up to 12 months from date of receipt. Avoid repeated freeze/thaw cycles.

Analysis Note

Retention of native structure was confirmed by ability to form microfibrils.
