Specificity

Uncoupling Protein 2 (UCP2), human. Human UCP2 is a 309 amino acid mitochondrial uncoupling protein (Fleury et al. 1997; Gimeon et al. 1997). It is only 59% homologous with UCP1 found in brown adipose tissues. UCP2 has wide tissue distribution in tissues (brain, kidney, liver, brown adipose tissue, heart, and muscle). UCP2 may play a critical role in energy balance, body weight, and thermoregulation (Fleury et al. 1997; Gimeon et al. 1997; Flier & Lowell 1997).

SPECIES REACTIVITIES: The antibody has not yet been tested for cross-reactivity to other species. The sequence used for immunization shows 100% homology to mouse and rat UCP2 and 92% homology with canine, porcine and bovine.

Immunogen

13 aa peptide sequence near the 3rd and 4th transmembrane domain of human UCP2 (Fleury et al. 1997; Gimeon et al. 1997). The peptide has no significant homology with UCP1, UCP3 or UCP4 (Boss et al. 1997).

Application

Detect Uncoupling Protein 2 using this Anti-Uncoupling Protein 2 Antibody validated for use in ELISA & WB.

Western Blotting: 1-10 μg/mL using Chemiluminescence technique.

Immunohistochemistry: Has not been tested. It is recommended that you use 2-20 μg/mL on formaldehyde fixed tissue.

ELISA: 1:10,000-1:100,000; using 50-100 ng UCP2 control peptide/well.

Optimal working dilutions must be determined by end user.

Doxorubicin-based chemotherapy is limited by the development of dose-dependent left ventricular dysfunction and congestive heart failure caused by reactive oxygen species (ROS). Uncoupling proteins (UCP) can inhibit mitochondrial ROS production as well as decrease myocyte damage from exogenous ROS. Prior
