General description

Lactate is generated by lactate dehydrogenase (LDH) under hypoxic or anaerobic conditions. Monitoring lactate levels is, therefore, a good indicator of the balance between tissue oxygen demand and utilization and is useful when studying cellular and animal physiology.

The kit is suitable for the quantitative determination of L-lactate (L-lactic acid) and the evaluation of drug effects on its metabolism in serum, plasma, urine, cell culture media, and other biological samples.

Biochem/physiol Actions

The L-Lactate Assay Kit is based on the lactate dehydrogenase catalyzed oxidation of lactate, in which the formed NADH reduces a probe into a highly fluorescent product. The fluorescence intensity of this product, measured at λEx = 530 nm/λEm = 585 nm, is proportional to the lactate concentration in the sample.

  1. Could you provide information on the measurement uncertainty?

    1. Specific information on the measurement uncertainty of this product is not available; however, during the development of these kits, an error of less than 10% is targeted, with all measurements performed in duplicate. Therefore, a fairly low measurement of uncertainty can generally be expected when running tests. The standard curve is also run with every group of samples to normalize results and obtain more consistent data.

  2. The kit contains a probe, but no instructions in the protocol of when/how much to add. Please advise on the probe-adding step?

    1. The protocol indicates that 5 uL of the probe should be added to the working reagent. This is described in Table 2, located on page 3 of the datasheet. Please see the link below to review:
