Application

Hydrocortisone has been used as a supplement in culture media for oligodendrocytes, colon cells and epithelial cells.[1]

Biochem/physiol Actions

Primary glucocorticoid secreted by the adrenal cortex.
Primary glucocorticoid secreted by the adrenal cortex. It has three times the anti-inflammatory potency of corticosterone but much lower Na2+ retention potency.
Hydrocortisone (also referred to as cortisol) is an endogenous glucocorticoid which is secreted by the adrenal gland. It is produced from the hypothalamus–pituitary–adrenal (HPA) axis. Hydrocortisone interacts with glucocorticoid receptors and regulates various signaling pathways. It is associated with the induction of energy mobilization, increase in cerebral perfusion rates and local glucose consumption. It is also linked with increase in cardiovascular output and respiration, redistribution of blood, delivery of molecules and energy to the brain and muscles and modulation of immune responses.

Preparation Note

To prepare 50 μg/mL stock solution, add 1.0 mL absolute ethanol to 1.0 mg of product, gently swirl to dissolve, add 19 mL sterile medium.

  1. How long can the powder be stored at RT? Reconstituting 1000mg to obtain 50mg/mL calls for 20mL of chloroform/ethanol, I need 100 aliquots of 50mg/ul in 10uL, therefore 20mL is too much, can I use only 50mg and save the rest for later use?

    1. This product is considered stable but may degrade over time. A periodic retest date is assigned to confirm quality, and the lot-specific retest date is reported on the Certificate of Analysis. It is acceptable to weigh out only the required amount of hydrocortisone powder (e.g., 50 mg), dissolve it in the appropriate volume of solvent, and save the remaining powder for later use. The remaining powder should be stored tightly closed, protected from moisture and light, and at room temperature.

  2. How long can you keep the powder stored at 4°C without a negative impact on effectiveness?

    1. This product is typically stored at room temperature. Stability studies at 4°C have not been conducted. However, storing the material at refrigerated temperatures is not expected to adversely effect the activity. Upon removal from the refrigerator, the bottle should be allowed to equilibrate prior to opening to avoid condensation and moisture uptake.

  3. How long is the stock solution reconstituted in absolute ethanol stable for at room temperature and 4C?

    1. Stock solutions are most stable when stored in aliquots at -20°C, protected from light. Solutions are expected to remain stable for 2 - 4 weeks when refrigerated at 2-8°C. The stability of this product in solution at room temperature has not been evaluated. Commercially available products, such as Solu Cortef® solutions, stored at room temperature at >2 mg/mL, are recommended to be discarded after 3 days or sooner if a change in color or turbidity is noted.

  4. Can this compound be dissolved in DMSO, aliquoted to avoid the freeze thaw cycle, and store at -20C instead of chloroform:ethanol mixture?

    1. The solubility of this product in DMSO has not been determined. However, various sources state that this product is soluble in DMSO at a minimum of 20 mg/mL.

  5. How long can the powder be stored after date of receipt?

    1. This product, while considered stable, may be subject to degradation over time and is assigned a periodic retest date to confirm quality.   6. How stable is hydrocortisone?  I use it and supplement a medium but I administer it every 48 hours, can I have my stock for 7 days and it does not lose its effect or do I have to defrost it every time I use it so that it does not lose its effect?

    1. The stability of this product in solution has not been determined. To assure optimum performance, it is recommended that solutions be stored in frozen aliquots at -20°C. However, the water soluble version of Hydrocortisone, available as product H0396, is estimated to be stable for 1 - 2 weeks when refrigerated at 2 - 8°C. Note that a publication, J. AM. PHARM. ASSOC., 49, 253 (1960), determined that solutions prepared in ethanol at 40mg/mL and stored at 25°C decline by 50% in potency after 160 days.

  7. Can it be dissolved completely in absolute ethanol, and stored per instructions (instead of 1:1 chloroform and ethanol)?

    1. A stock solution can be prepared by reconstituting the product at 1 mg/ml in 100% EtOH. Gently swirl to dissolve. Then dilute to 50 ug/ml with sterile culture media, aliquot, and store at -20 °C. As a frozen aliquot, this should be stable for at least 3-4 months. Avoid the freeze/thaw cycle.

    hard to aliquot

    the powder sticks to a lot of surfaces and it is hard to aliquot it,
    it should be solubilized in water with ethanol
