Cell Line Description
tsA201 is a transformed human kidney (293, Sigma catalogue no. 85120602) cell line stably expressing an SV40 temperature-sensitive T antigen. The cell line has been used in a variety of functional expression assays and has been reported to produce high levels of recombinant proteins. Transfection efficiency decreases over a period of several months, therefore it is advisable to lay down a frozen stock as a reserve. The cells have been eradicated from mycoplasma at ECACC. The identity of tsA201 and 293 has been confirmed by STR profiling.
Application
tsa201 human cell line has been used for protein-binding studies, whole-cell patch clamp recordings and transfection studies.

Culture Medium
DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Subculture Routine
Split sub-confluent cultures (70-80%) 1:2 to 1:8; cells are dislodged by mechanical agitation or EDTA solution; 5% CO2; 37°C. Do not let cells become over confluent as this can affect morphology.
