General description

The DNase Test Agar with Toluidine Blue is a specialized microbiological medium used to detect deoxyribonuclease (DNase) activity in bacteria and fungi. It incorporates toluidine blue as an indicator, which produces a distinct color change when DNase activity is present. This medium is particularly effective for differentiating and identifying nonpigmented Serratia species. Tryptose in the formulation provides essential nutrients, while DNase activity depolymerizes DNA, triggering a bright pink reaction due to the metachromatic properties of toluidine blue. However, some Staphylococcus strains may experience growth inhibition on this medium because of the toluidine blue. Additional confirmatory tests are recommended for accurate identification.

Application

DNase Test Agar with Toluidine Blue is recommended for the detection of deoxyribonuclease activity of bacteria and fungi, especially for the identification of pathogenic Staphylococci. This media can be used in food and beverage testing, clinical microbiology, and general microbiological research.

Preparation Note

Suspend 42.1 g of DNase Test Agar with Toluidine Blue in 1000 ml of distilled water. Heat with frequent agitation to dissolve the medium completely. Sterilize by autoclaving at 12 to 15 lbs. pressure (118°C to 121°C) for 15 minutes. Cool to 45°C and pour into sterile petri plates.
