General description

m-FC Agar is a selective medium designed for the detection and enumeration of fecal coliform bacteria in water samples. It is widely used in environmental microbiology for water quality testing. The medium contains selective agents that inhibit the growth of non-target organisms while promoting the growth of fecal coliforms. Proteose peptone, Tryptose, and Yeast extract act as sources of carbon, nitrogen, minerals, vitamins, and other essential growth nutrients. Lactose is a fermentable carbohydrate. mFC Agar contains Bile salts, which inhibit the accompanying gram-positive bacteria. Methyl blue is a triphenylmethane dye that suppresses the growth of many gram-positive bacteria. The indicator system is based on the color change of methyl blue and rosolic acid. The high incubation temperature makes the test more selective.

m-FC Agar is used for the detection and enumeration of fecal coliforms using membrane filter technique. This medium is suitable for detecting fecal coliforms in water samples, ensuring compliance with environmental and public health standards.

Preparation Note

Dissolve 52 g in 1 litre distilled water and heat to boiling to dissolve completely. Add 10 ml of a 1 % solution of rosolic acid (Cat. No. 861324) in 0.2 N NaOH. Continue heating for 1 minute with frequent agitation. Do not autoclave! Cool to 45-50°C. Dispense 4 ml amounts into Petri dishes (50-60mm) and allow to solidify.
