General description
Acetamide Nutrient Broth is a synthetic basal mineral medium designed for the detection of microbial utilization of acetamide by a wide variety of non-fermenting organisms. Acetamide serves as the sole substrate providing both nitrogen and carbon sources, while potassium phosphate acts as the buffering agent, and other inorganic salts supply essential minerals required for microbial growth. Organisms growing in this medium metabolize acetamide through acylamidase activity, a characteristic that has been detected in Alcaligenes odorans, Pseudomonas aeruginosa, and Pseudomonas acidovorans group III (Achromobacter xylosoxidans). It is important to note that some strains deaminate acetamide slowly and may require up to 7 days of incubation, and only approximately 40% of pyocyanogenic strains of Pseudomonas aeruginosa will show a positive reaction. Therefore, additional confirmatory tests are recommended when results are negative to ensure accurate identification.
Application
Acetamide Nutrient Broth is used for the detection and identification of non-fermenting microorganisms capable of utilizing acetamide as a sole carbon and nitrogen source. The medium is used in environmental and water microbiology laboratories for the presumptive identification of Pseudomonas aeruginosa and related non-fermenting organisms, where the ability to deaminate acetamide serves as a key biochemical marker for differentiation and identification purposes.

Preparation Note
Directions:
Dissolve 0.56 g Acetamide Nutrient Broth Part A and 2 g Acetamide in 950 ml distilled water. Check the pH and if necessary, adjust. Make up to 1 litre and pour into tubes. Autoclave at 121°C for 15 minutes.
Technique:
Medium is inoculated with a couple off loops from the culture to be assayed and it is incubated at 32-35°C for 24-48 hours before going to the isolation medium.
To confirm Ps. aeruginosa, inoculate a loop of culture in Asparagine Proline Broth (Cat. No. 17129) and incubate at 35-37°C for 24 hours. After this period, pour 1-2 drops of Nessler′s Reagent
(Cat. No. 72190) on to the culture and verify ammonia production: a change to yellow will mean ammonia production positive and therefore, Ps. aeruginosa presence.
