Selectins (CD62s) are single-chain transmembrane glycoproteins that function as cell adhesion molecules (CAMs) by interacting with a range of oligosaccharide structures including sialyl-Lewis X (sLeX) via their C-type lectin domain. All three known selectins (L-, E-, and P-selectin or CD62L CD62E, CD62P, respectively) share a similar overall domain structure that includes an N-terminal C-type lectin domain (calcium-dependent lectin domain), an EGF-like domain, a variable number of short consensus repeat (SCR) or Sushi domains (2, 6, and 9 for CD62L CD62E, CD62P, respectively), a transmembrane domain, and a cytoplasmic tail. The extracellular domains show a high homology among the three selectins, the transmembrane and cytoplasmic domains however are not conserved among the three. E-selectin (CD62E, ELAM-1) and P-selectin (CD62P, GMP-140, LECAM3, PADGEM) are mainly expressed on endothelial cells at inflammatory sites, whereas L-selectin (CD62L) is exclusively expressed on leukocytes.

Biochem/physiol Actions
This antibody is specific to human antigen E-selectin (ELAM-1, 115 kDa) and P-selectin that are expressed on activated endothelial cells. The ligand is expressed on neutrophils and may function as an early part in neutrophil binding to the endothelium. This antibody inhibits neutrophil adhesion to E-selectin and P-selectin when used as F(ab′)2 fragments. Although previously thought to recognize only human CD62E, later data (Goda, K., et al., (1999). Eur. J. Immunol. 29(5):1551-1560) showed that this antibody also recognizes human CD62P by binding to a common epitope on these members of the selectin family.
