General description

A cell-permeable, non-fluorescent, and hydrophobic compound, which is rapidly hydrolyzed by intracellular esterases releasing the membrane-impermeable, hydrophilic, and intensely fluorescent calcein. Can also be used for imaging mitochondrial permeability pore transition. Calcein is better retained by viable cells than fluorescein, carboxyfluorescein, or BCECF, and its fluorescence is brighter in a variety of mammalian cell types. Useful for assessing adhesion, cell viability, chemotaxis, and multidrug resistance (MDR).
A cell-permeable, nonfluorescent, and hydrophobic compound, which is rapidly hydrolyzed by cytoplasmic esterases inside the cell, releasing the membrane-impermeable, hydrophilic, and intensely fluorescent calcein (excitation maximum: 494 nm; emission maximum: 517 nm). A cytosolic fluorophore that can also be used imaging mitochondrial permeability pore transition. Calcein is better retained by viable cells than fluorescein, carboxyfluorescein, and BCECF and its fluorescence is brighter in a variety of mammalian cell types. Useful for assessing adhesion, cell viability, chemotaxis and multidrug resistance (MDR).

  1. Good morning, is it possible to use CALCEIN-AM alone diluited in DMSO for cells observation on scaffolds? In that case, would it be enough to diluite the product directly in DMSO at a concentration of 5 mg/mL? Thank you in advance

    1. Calcein-AM is soluble directly in anhydrous DMSO at 5 mg/mL. However, the stock solution is typically diluted to a range of 0.1-10 µM in buffer or medium for cell staining. It is not recommended to add the DMSO stock solution directly to cells.

    1. This product stored at -20°C, but ships at ambient temperature. When there may be short-term exposure (i.e. during shipment) to conditions other than those recommended for long-term storage that will not affect product quality, Sigma-Aldrich will ship at ambient temperature.
