General description
NSC697923 stimulates apoptosis in p53 mutant neuroblastoma (NB) cells by triggering JNK (c-Jun N-terminal kinases) pathway. This ubiquitin-conjugating enzyme E2 N (UBE2N) inhibitor is considered as an important treatment option for neuroblastoma (NB) patients. NSC697923 prevents the multiplication and survival of diffuse large B-cell lymphoma (DLBCL) cells in vitro.
Application
NSC697923 has been used as an inhibitor in STING-dependent NF-κB (nuclear factor-κB) activation after DNA damage, mediated by TRAF6 (TNF receptor-associated factor 6).
Biochem/physiol Actions
NSC697923 is a cell-permeable and selective inhibitor of the ubiquitin-conjugating (E2) enzyme Ubc13-Uev1A that blocks the formation of the Ubc13 - ubiquitin thioester conjugate, and suppresses constitutive NF-kB activity in ABC-DLBCL cells. Also, NSC697923 inhibits the proliferation and survival of ABC-DLBCL and GCB-DLBCL cancer cells.

Neuroblastoma (NB) is the most common extracranial neoplasm in children. In NB, loss of p53 function is largely due to cytoplasmic sequestration rather than mutation. Ubiquitin-conjugating enzyme E2 N (UBE2N), also known as Ubc13, is an E2 ubiquitin-conjugating enzyme that

Ubc13 is an E2 ubiquitin conjugating enzyme that functions in nuclear DNA damage signaling and cytoplasmic NF-κB signaling. Here, we present the structures of complexes of Ubc13 with two inhibitors, NSC697923 and BAY 11-7082, which inhibit DNA damage and NF-κB

Protein ubiquitination has been historically associated with protein degradation, but recent studies have demonstrated other cellular functions associated with substrate ubiquitination. Among the RING-type ubiquitin E3 ligase enzymes present in the human genome, RNF167 is a transmembrane protein located in
