ATAD3A and ATAD3B (MIM 612317) are mitochondrial membrane proteins that contribute to the stabilization of large mitochondrial DNA (mtDNA)-protein complexes called nucleoids (He et al., 2007 [PubMed 17210950]).[supplied by OMIM]
ATPase family, AAA domain containing 3A (ATAD3A) is a mitochondrial protein, encoded by the gene mapped to human chromosome 1p36.33. The encoded protein is one of the paralogs of ATAD3 gene family. ATAD3A is ubiquitously expressed.

Biochem/physiol Actions

ATPase family, AAA domain containing 3A (ATAD3A) regulates mitochondrial dynamics and aids in the interaction between mitochondria and the endoplasmic reticulum. In addition, the encoded protein is also involved in mitochondrial development, organization of nucleoid, translation of protein, cell growth and cholesterol metabolism. Elevated expression of ATAD3A serves as a biomarker for radioresistance in glioblastoma multiforme. It is also associated with cancers such as uterine cervical cancer, lung adenocarcinoma and prostate cancer. Mutation in ATAD3A causes delayed development, optic atrophy, axonal neuropathy, hypotonia and hypertrophic cardiomyopathy. Silencing ATAD3A expression promotes autophagy and apoptosis in uterine cervical cancer cells and reduces resistance to anticancer drugs.
