General description

Ubiquitin specific peptidase 51 (USP51) encodes a deubiquitylating enzyme. USP51 is predominantly expressed in prostate, brain, lung, aorta and kidney. In human chromosome, the gene USP51 is localized on Xp11.

Biochem/physiol Actions

Ubiquitin specific peptidase 51 (USP51) in response to DNA damage deubiquitylates histone H2A at Lys13,15 (H2AK13,15ub) present at DNA double-strand breaks (DSBs) to maintain genome integrity. High expression of USP51 suppresses ionizing radiations induced damage on p53-binding protein 1(53BP1) and breast cancer type 1 susceptibility protein (BRCA1). Cells depleted with USP51 show an increase in G1 phase cells and decrease in S-phase cells and hence it is required for cell cycle progression. USP51 deubiquitinase zinc finger E-Box binding homeobox 1)(ZEB1) and stabilizes its expression, especially in breast tumour cells.
