General description
Anti-Smac/DIABLO is developed in rabbit using a synthetic peptide corresponding to the C-terminus of human Smac/DIABLO-L. Second mitochondria-derived activator of caspase/direct inhibitors of apoptosis protein (IAP) binding protein with low pI (Smac/DIABLO) is a mitochondria-derived pro-apoptotic protein that is released from the mitochondria in response to apoptotic stimuli. It exists as two isoforms, Smac/DIABLO-L and a short isoform Smac/DIABLO-S, differing in their N-terminal sequences. Smac/DIABLO-L also contains an N-terminal signal sequence. It is highly expressed in adult testis, ovary, liver, kidney, spleen, prostate and heart. This gene is located on human chromosome 12q24.31.

Application
Anti-SMAC/Diablo antibody produced in rabbit may be used in immunoblotting and immunohistochemistry.
Biochem/physiol Actions
Second mitochondria-derived activator of caspase/direct IAP binding protein with low pI (Smac/DIABLO) is a proapoptotic protein, which promotes the caspase activity of the initiator caspase 9 and the effectors caspase 3 and caspase-7 by neutralizing the inhibitory activity of the inhibitors of apoptosis proteins (IAPs).It promotes apoptosis by removing XIAP from the apoptotic protease activating factor-1 (Apaf-1)/caspase-9 apoptosome. The N-terminal AVPIA sequence in mature Smac/DIABLO is absolutely required for interaction with the baculovirus IAP repeat 3 (BIR3) domain of XIAP. Mutations that disrupt Smac/DIABLO dimer formation abrogate the XIAP-neutralizing activity of this molecule, suggesting that Smac/DIABLO dimerization is essential for its proapoptotic activity. It plays a key role intrinsic and extrinsic in apoptotic pathways. Smac mRNA is overexpressed in de novo acute leukemia (AL) patients.
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, and 15 mM sodium azide.
Storage and Stability
For continuous use, store at 2-8 °C for up to one month.For prolonged storage, freeze in working aliquots at−20 °C. Repeated freezing and thawing is not recom-mended. Storage in frost-free freezers is also notrecommended. If slight turbidity occurs upon prolongedstorage, clarify the solution by centrifugation beforeuse. Working dilutions should be discarded if not usedwithin 12 hours.
